The discovery of the Yamanaka factors allowed scientists to induce pluripotency in a way that was unachievable before, but the implementation proved to be complex. Previous methods in cellular reprogramming often create problems with temporal control of the reprogramming genes, site-specific delivery, and insertional mutagenesis. One of the methods used to control gene expression with adenovirus is the Tet-On system, which is only able to regulate the reprogramming genes in a limited manner through a controlled system. If we substitute the reprogramming genes of the tet-on system in the adenovirus with small activating RNAs (saRNAs), the coding regions of the virus could be expressed for a shorter amount of time and still achieve the desired effect. Furthermore, the saRNAs would target the promoter regions of the reprogramming factors, allowing for a more efficient and controllable reprogramming process. As a result, replacing reprogramming genes with saRNAs would effectively overcome the limitations of previous methods. This would allow researchers to reprogram cells with greater efficiency and produce safer induced pluripotent stem cells (iPSCs) for cell or tissue transplants and treating a myriad of diseases such as spinal cord injuries, heart diseases, leukemia, Alzheimer’s, and more.
