      Bamboo charcoal mediated plant secondary metabolites biosynthesis in tomato against South American tomato pinworm (Tuta absoluta)

          Abstract

          Introduction

          The south American tomato pinworm (Tuta absoluta), an invasive insect pest species, has established itself in more than 33 Chinese prefectures, where it is mainly damaging tomato crops. Immediate efforts have been initiated to find strategies to control this pest. Studies have shown that biochar (BC) amendment to soil can enhance plant growth and resistance to herbivory.

          Methods

          First, we quantified the morphological performance of tomato plants grown in different coconut bran and bamboo charcoal (v/v) combinations and selected the most beneficial one. Then we checked the T. absoluta survival on the tomato plants grown in the selected bamboo charcoal combination. Finally, we studied the leaf metabolite accumulation and gene expression changes in tomato plants after growing in the selected bamboo charcoal combination.

          Results

          We found that the 30:1 ratio of bamboo charcoal and coconut bran is the most beneficial to tomato growth as its amendment to soil increased tomato plant height, stem thickness, and chlorophyll content, whereas, the T. absoluta survival decreased. The metabolome profiles of BC tomato leaves showed an increased accumulation of flavonoids, terpenoids, and phenolic acids compared to CK. Transcriptome sequencing resulted in the identification of 244 differentially expressed genes. Most of the upregulated genes were associated with stress-related hub proteins, flavonoid biosynthesis, MAPK and phytohormone signaling, and terpenoid biosynthesis. Additionally, the expression of many genes related to signaling and defense was changed in response to the bamboo charcoal amendment.

          Discussion

          We conclude that bamboo charcoal induces biosynthesis of flavonoids, terpenoids, and phenolic acids, which improve plant growth and tolerance against T. absoluta, thus reducing the survival of destructive pests.

          Plant immunity to insect herbivores.

          Herbivorous insects use diverse feeding strategies to obtain nutrients from their host plants. Rather than acting as passive victims in these interactions, plants respond to herbivory with the production of toxins and defensive proteins that target physiological processes in the insect. Herbivore-challenged plants also emit volatiles that attract insect predators and bolster resistance to future threats. This highly dynamic form of immunity is initiated by the recognition of insect oral secretions and signals from injured plant cells. These initial cues are transmitted within the plant by signal transduction pathways that include calcium ion fluxes, phosphorylation cascades, and, in particular, the jasmonate pathway, which plays a central and conserved role in promoting resistance to a broad spectrum of insects. A detailed understanding of plant immunity to arthropod herbivores will provide new insights into basic mechanisms of chemical communication and plant-animal coevolution and may also facilitate new approaches to crop protection and improvement.

            Plant Flavonoids—Biosynthesis, Transport and Involvement in Stress Responses

            This paper aims at analysing the synthesis of flavonoids, their import and export in plant cell compartments, as well as their involvement in the response to stress, with particular reference to grapevine (Vitis vinifera L.). A multidrug and toxic compound extrusion (MATE) as well as ABC transporters have been demonstrated in the tonoplast of grape berry, where they perform a flavonoid transport. The involvement of a glutathione S-transferase (GST) gene has also been inferred. Recently, a putative flavonoid carrier, similar to mammalian bilitranslocase (BTL), has been identified in both grape berry skin and pulp. In skin the pattern of BTL expression increases from véraison to harvest, while in the pulp its expression reaches the maximum at the early ripening stage. Moreover, the presence of BTL in vascular bundles suggests its participation in long distance transport of flavonoids. In addition, the presence of a vesicular trafficking in plants responsible for flavonoid transport is discussed. Finally, the involvement of flavonoids in the response to stress is described.

              Ecology, Worldwide Spread, and Management of the Invasive South American Tomato Pinworm, Tuta absoluta: Past, Present, and Future.

              The South American tomato pinworm, Tuta absoluta (Meyrick), is native to the eastern Neotropics. After invading Spain in 2006, it spread rapidly throughout Afro-Eurasia and has become a major threat to world tomato production. Integrated pest management (IPM) strategies have been developed, but widespread insecticide use has caused selection for insecticide resistances as well as undesirable effects on key beneficial arthropods. Augmentation and conservation biological control relying on omnivorous mirid predators has proved successful for management of T. absoluta, where implementation is dependent on abiotic, biotic (e.g., alternative prey), and anthropogenic factors (e.g., pesticides). Research has been carried out on larval parasitoids, showing potential for further development of sustainable control. The development of resistant tomato varieties is ongoing, but they are not commercially available yet. Knowledge gaps remain to be filled to optimize IPM packages on tomato crops and to help prevent further spread worldwide.
